Abstract: The Drug Repurposing Hub, established by the Broad Institute of MIT and Harvard, stands as a beacon of open-access resources in the realm of drug discovery. Featuring a meticulously curated assortment of globally sanctioned, and preclinical drugs and compounds, accompanied by comprehensive annotations, this repository boasts over 6,800 small molecules—one of the most comprehensive in the world. Traditionally employed to assess against disease models, with the aspiration of unearthing novel applications for these finely tuned compounds, the Hub now serves as a catalyst for unveiling fresh perspectives on disease mechanisms. In tandem with the Center for the Development of Therapeutics (CDoT) at the Broad Institute, this unique collection is utilized by scientists in their high-throughput screening campaigns as a “molecular toolbox.” CDoT leverages this richly annotated repository in high-throughput screening pursuits, translating curiosity-driven academic research to identify hit candidates and unbiasedly uncover and explore new pathways affecting the biology of interests. While the repository undeniably holds promise in unveiling new applications for established drugs, its true potential is unlocked when applied to discover new biological insights and disease mechanisms.
