Abstract: The TruSight Oncology 500 is a next-generation sequencing (NGS) assay with a comprehensive genomic profiling of tumor samples that can assess over 500 genes of DNA and RNA variant types, plus microsatellite instability (MSI) and tumor mutation burden (TMB). A timely and efficient workflow is essential to producing reliable sequencing samples. The goal was to evaluate a high-throughput solution that reduces the chance for human errors and minimizes hands-on-time. Here we discuss the use of the Revvity Fontus NGS workstation to generate NGS libraries with the TruSight Oncology 500 workflow. The performance was evaluated using human reference DNA and RNA through the automated workflow. The final library yields, and size distribution were reproducible and consistent across the plate, producing sequencing libraries within the range of the kit. The automated workflow offers a streamlined solution that can process up to 96 samples, increasing lab productivity, and providing consistent performance.
