(1138-A) Leveraging Evotec Sample Management’s Expertise and Versatility for Efficient Integrated Research Projects: from Compound Acquisition to Biological Insights.

Abstract: Evotec Toulouse Sample Management (SM) is a state-of-the-art facility which supports drug discovery on small chemical molecules and biologics. With cutting-edge technologies, it offers a wide range of services, from secure storage of compounds to reformatting and delivery across the world in a timely manner. In this poster we present a case study demonstrating the versatility and pivotal role of Evotec’s SM at the service of efficient and integrated drug discovery projects for our partners. 
Evotec’s experts in compound sourcing acquired a 5k library on behalf of a partner according to its very specific criteria. Various commercial compound suppliers were evaluated and carefully selected. Samples were directly shipped to the Toulouse SM facility. Data was registered in the SM LIMS (Mosaic®) and in Dotmatics database. Within a week, these 5k compounds were reformatted using a Beckman I7 pipettor from 96 Matrix tube racks to 8 assay plates with various reference, layout, volume and concentration. A dedicated SM - QC team in Toulouse performed a high throughput analysis of these 5k compounds by LC/MS to validate identity and purity, using the Analytical StudioTM software. Assay plates were then shipped across 3 Evotec sites (UK, Italy, US)  to be tested in dedicated ADME-Tox assays. Various physicochemical and biological properties of molecules were evaluated, such as solubility value, hydrophobicity index, hERG inhibition or permeability. Remaining samples were securely stored in a -20°C Azenta fully automated store in Toulouse.
After successfully processing this first set of compounds, a regular shipment with a same day turnaround time of ~30 project samples was set up twice a month with a CRO in India. Evotec logistics team ensured shipment and customs forms processing using a dedicated broker to reduce delivery timelines. In this process, SM receives, register, solubilizes, plate-out and ships back these compounds within the same day.
Fast and reliable compound management is essential to reduce cycle times in modern drug discovery. Thanks to optimized complete workflows, Evotec’s SM demonstrates its capability to be an efficient actor in this field.
