Abstract: The ability to obtain and map transcripts from individual cells has greatly enhanced our understanding of life processes and disease mechanisms. Scale Biosciences’ Single Cell RNA Sequencing Kit enables single-cell analysis by using the cell itself as the reaction compartment. Highly parallelized barcode combinations scale exponentially, facilitating higher sample and cell throughput.
While these workflows have improved accessibility significantly without the need for specialized instruments, automation can further streamline processing time while reducing user pipetting errors and strain. As a plate-based technology, ScaleBio’s method integrates seamlessly with automated liquid handling systems, offering ease of use and reduced hands-on time.
In this poster, we present an automated workflow for the ScaleBio Single Cell RNA Sequencing Kit protocol using SPT Labtech’s all-in-one liquid handling platform, firefly®. The combination of these technologies results in a streamlined workflow that enhances efficiency without compromising results. Both manually prepared libraries and those generated with firefly show comparable metrics, demonstrating that automation shows no detrimental effects to library quality and cell recovery.
An additional benefit is the significant reduction in consumables usage compared to traditional air displacement pipettors, achieved through firefly’s non-contact positive displacement reagent dispense head.
