Abstract: GPCRs (G Protein Coupled Receptors) represent the largest family of cell surface receptors and are the main target for drugs available on the pharmaceutical market. Luminescence-based techniques such as fluorescence, bioluminescence, FRET & BRET have been widely used to study GPCRs by monitoring their signaling pathways, including G-protein activation, recruitment of GRKs & arrestins, interaction with effectors and second messengers production. In the present work, we illustrate some fluorescence & BRET applications performed on the ARPEGE facility using the FDSS/μCELL imaging plate reader (Hamamatsu Photonics) enabling simultaneous real-time monitoring of signaling pathway kinetics in 96 wells at a time, providing valuable insights into signaling profiles.
